ABSTRACT

Abstract Aims: To identify the histopathological alterations in organs of Wistar rats to evaluate toxic effects of use of Annonamuricata Raw Leaf Extract (AMRLE) alone or in association with DMBA. Settings and Design: Sixty female Wistar rats were used, separated into groups and treated with a single dose of 65 mg/kg of DMBA and/or with 50; 100 and 200 mg/kg of AMRLE. Hematoxylin-Eosin (HE) and 1% methylene blue stains were used in the histopathological analysis and quantification of Aberrant Crypts (ACs) and Aberrant Crypt Focus (ACF). Fischer and Kruskal Wallis tests were used in the statistical analysis. Results: The administration of 65 mg/kg of DMBA and/or 50, 100 and 200 mg/kg of AMRLE did not influence weight development. Some histopathological alterations (hepatic steatosis; inflammatory foci in the liver, kidney and lung; pulmonary lymphoid hyperplasia, ectasia and hyperplasia in mammary gland epithelium) and the development of ACs and ACF in the intestinal colon were observed in all groups, except in the group negative control, with no statistical difference between analysed groups. Conclusions: Histopathological alterations and the formation of ACs and ACF did not show a statistically significant difference between the groups analysed. However, although AMRLE has antioxidant effects due to the presence of phenolic components, there was still the formation of some pathological processes that may be related to the isolated toxic action of DMBA and/or associated with other components of AMRLE, since these changes were not seen in the negative control group.


Resumen Objetivos: Identificar las alteraciones histopatológicas en órganos de ratas Wistar para evaluar los efectos tóxicos del uso del Extracto de Hoja Cruda de Annona muricata (AMRLE) solo o en asociación con DMBA. Configuración y diseño: Se utilizaron sesenta ratas hembras Wistar, se separaron en grupos y se trataron con una dosis única de 65 mg/kg de DMBA y/o con 50, 100 y 200 mg/kg de AMRLE. Se utilizaron tinciones de hematoxilina-eosina (HE) y azul de metileno al 1% en el análisis histopatológico y la cuantificación de criptas aberrantes (CA) y focus de criptas aberrantes (FCA). En el análisis estadístico se utilizaron las pruebas de Fischer y Kruskal Wallis. Resultados: La administración de 65 mg/kg de DMBA y/o 50, 100 y 200 mg/kg de AMRLE no influyó en el desarrollo del peso. Se observaron algunas alteraciones histopatológicas (esteatosis hepática; focus inflamatórios en el hígado, riñón y pulmón; hiperplasia, ectasia en epitelio de la glándula mamaria e hiperplasia linfoide pulmonar) y el desarrollo de CA y FCA en el colon intestinal en todos los grupos, excepto en el grupo control negativo, sin diferencias estadísticas entre los grupos analizados. Conclusiones: Las alteraciones histopatológicas y la formación de CA y FCA no mostraron diferencias estadísticamente significativas entre los grupos analizados. Sin embargo, aunque AMRLE tiene efectos antioxidantes debido a la presencia de componentes fenólicos, aún existe la formación de algunos procesos patológicos que pueden estar relacionados con la acción tóxica aislada del DMBA y/o asociados con otros componentes de AMRLE, ya que estos cambios no fueron observados en el grupo control negativo.

Resumen El objetivo de este estudio fue demostrar que los principios activos de las semillas de Annona muricata combinados con el extracto etanólico y dimetilsulfóxido (EE-DMSO), incrementan la mortalidad de larvas IV y pupas de Aedes aegypti con relación a extractos acuosos (EA) y extractos etanólicos (EE). Las bioactividades se calcularon por comparación de los porcentajes de mortalidad a las 6, 12, 24 y 48 horas in vitro y campo simulado. Los resultados indicaron mortalidad progresiva dependiente de las concentraciones y tiempos de exposición en larvas y reacción knock-down en pupas. In vitro a 5 mg.L-1, EA y EE ejercieron 100% de mortalidad larvaria en 24 horas de exposición (CL50=46.16 y 19.28 mg.L-1 respectivamente), en contraste con EE-DMSO, que inició sobre 62% con 0.5 mg.L-1 a las 6 horas (CL50=20.33 mg.L-1). La acción pupicida de EA y EE reveló 100% de mortalidad desde 24 horas en todas las concentraciones, a diferencia de EE-DMSO que se alcanzó entre 6 y 12 horas. En campo simulado, EA y EE ejercieron 100% de mortalidad a las 24 horas en larvas (16.91 y 21.21 mg.L-1 ), mientras que en pupas (20.44 y 23.03 mg.L-1) ocurrió a las 12 horas, entretanto, la actividad pupicida de EE-DMSO fue 100% en 6 horas. Los efectos comparativos in vitro y campo simulado denotaron patrones similares de respuestas larvicida y pupicida, pero con mayor sensibilidad en pupas. Los principios activos de las semillas de A. muricata combinados con EE-DMSO potencian la respuesta mortal de larvas y pupas de A. aegypti in vitro y campo simulado.


Abstract The objective of this study was to demonstrate that active ingredients of Annona muricata seeds can be enhanced as a result of mixture of both ethanolic extract of A. muricata seeds and Dimethylsulfoxide (EE-DMSO). Percentage mortalities at 6, 12, 24 and 48 hours on fourth instar larvae and pupae of Aedes aegypti were calculated in order to compare bioactivities of aqueous (AE), ethanolic extracts (EE) and EE-DMSO under laboratory and simulated field conditions. Results showed larval mortality concentration- and time-dependent, and knock-down responses in pupae. In laboratory, AE and EE exerted 100% larval mortality at 5 mg.L-1 after 24 hours (LC50= 46.16 and 19.28 mg.L-1). Conversely, EE-DMSO showed between 62 - 100% mortality at 0.5 mg.L-1 for over 6 hours (LC50= 20.33 mg.L-1). Pupicidal effects in AE and EE revealed 100% mortality at 24 hours employing all concentrations, except in EE-DMSO which commenced when individuals were exposed between 6 and 12 hours. In simulated field, AE and EE provoked 100% larval mortality at 24 hours (16.91 y 21.21 mg.L-1) while pupal mortality at 12 hours (20.44 y 23.03 mg.L-1). Percentage mortality of pupae was 100% using EE-DMSO even before 6 hours. Comparative toxic effects of laboratory and simulated-field systems have shown to maintain a similar pattern of larval mortality and more sensitive responses in pupae. Accordingly, larval and pupal mortality responses of A. aegypti were enhanced with the use of EE-DMSO and active ingredients of A. muricata seeds under laboratory and simulated field conditions.

ABSTRACT

O hidromel é uma bebida oriunda da fermentação alcoólica de mosto de mel e alguns adjuntos, como as frutas, para o fornecimento de nutrientes essenciais à fermentação. A graviola e o feijão caupi são importantes matérias-primas vegetais brasileiras, que constituem importantes fontes de carboidratos, proteínas, sais minerais e outros compostos. Assim, o presente trabalho teve como objetivo avaliar o efeito da suplementação de mostos de mel, com concentração fixa de feijão caupi (30 g.L-1) e diferentes concentrações de graviola (0, 10 e 30% m/m), sobre a fermentação dos mesmos, avaliando os perfis de concentração celular e dos teores de sólidos solúveis totais e etanol. Diante dos resultados obtidos foi possível observar que a graviola pode ser considerada um bom suplemento para o estímulo da fermentação de mostos de mel por leveduras.

ABSTRACT

Objective:To identify the phytochemical compounds from Annona muricata (A. muricata) and to determine their in vitro anti-proliferative activities against breast cancer cells, MCF7 and MDA-MB-231.Methods:A. muricata leaves were successively extracted by soxhlet method using n-hexane, ethyl acetate and methanol, and decocted with water. Each extract was analysed by gas chromatography mass spectrometry (GCMS) and characterized with Wiley and NIST library searches. Anti-proliferative activity of each extract was evaluated on MCF7 and MDA-MB-231 breast cancer cells using MTT assay.Results:The GCMS analysis of different solvent extracts of A. muricata leaves showed presence of different chemical groups of compounds such as steroids, terpenoids, phenolic compounds, sugars, sugars alcohol and others including vitamin E. Ethyl acetate leaves extract exhibited the lowest ICConclusion:Steroids and phenolic compounds were the main phytocompound groups identified from all A. muricata leaves extracts. The antiproliferative activity of n-hexane and ethyl acetate extract towards breast cancer MCF7 and MDA-MB-231 respectively might be due to the presence of biologically active compounds in the extracts, hence, providing some scientific evidences of the effectiveness of its traditional usages.

ABSTRACT Graviola leaves (Annona muricata L., Annonaceae) are used by some people to try to treat or even cure cancer, even though over-consumption of the fruit, which contains the neurotoxins annonacin and squamocin has caused an atypical form of Parkinson's disease. In previous analyses, the fruits were extracted with methanol under ambient conditions before analyses. In the present study, UPLC–QTOF–MS and NMR were used to analyze freeze-dried graviola leaves that were extracted using dry methanol and ethanol at 100 ºC and 10 MPa (100 atm) pressure in a sealed container. Methanol solubilized 33% of the metabolites in the lyophilized leaves. Ethanol solubilized 41% of metabolites in the lyophilized leaves. The concentrations of total phenolic compounds were 100.3 ± 2.8 and 93.2 ± 2.0 mg gallic acid equivalents per g of sample, for the methanolic and ethanolic extracts, respectively. Moreover, the toxicophore (unsaturated γ-lactone) that is present in neurotoxic acetogenins was found in the lipophilic portion of this extract. The concentrations of the neurotoxins annonacin and squamocin were found by UPLC–QTOF–MS to be 305.6 ± 28.3 and 17.4 ± 0.89 µg/g-dw, respectively, in the dried leaves. Pressurized methanol solubilized more annonacin and squamocin than ethanol. On the other hand, a hot, aqueous infusion solubilized only 0.213% of the annonacin and too little of the squamocin to be detected. So, graviola leaves contain significant amounts of the neurotoxins annonacin and squamocin, as well as some potentially healthy phenolic compounds. Finally, the potential neurotoxicity of whole leaves in dietary supplements could be much higher than that of a tea (hot aqueous infusion) that is made from them.

ABSTRACT

Annona muricata Linn has been reported to contain valuable bioactive compounds known as Annonaceous acetogenins.Theselong chain fatty acids were widely discussed for its potential in promoting anticancer and antiproliferative activity in various cancer cell lines. However, little study has been done on A. muricata effect in pancreatic cancer cells. In this study, the viability of Capan-1 after treatment with A. muricata extracts was determined by using 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. The results displayed that only hexane and commercialized extract inhibited cell proliferation in a concentration-dependent manner with IC25 varied ~7.8-8μg/ml and ~0.9-1.0μg/ml respectively. The data demonstrate that A. muricata hexane and commercialized extracts induced mild cytotoxicity in pancreatic cancer cells (Capan-1).

O presente trabalho objetivou avaliar o efeito da adição de inulina (I) e a substituição parcial da gordura do leite (G) pelo concentrado de proteína de soro de leite (WPC) sobre a sobrevivência dos probióticos Lactobacillus acidophilus NCFM e Bifidobacterium animalis subsp. lactis HN019 em sorvete de graviola com teor reduzido de gordura, ao longo do período de armazenamento e frente às condições encontradas no trato gastrointestinal (TGI) simuladas in vitro. Adicionalmente, avaliou-se a influência desses ingredientes (6% I; 1,5% WPC; 3% e 1,5% G) sobre as características tecnológicas e a aceitabilidade do sorvete funcional. Empregou-se um planejamento fatorial 22, para 4 formulações produzidas, em triplicata, totalizando 12 ensaios: F1- controle (- I, - WPC); F2 (+ I, - WPC); F3 (- I, + WPC) e F4 (+ I, + WPC). Todas as formulações foram armazenadas a -18±3ºC e avaliadas após 2, 28, 56, 84 e 112 dias de armazenamento. A determinação das características tecnológicas foi realizada com as análises de dureza instrumental (em analisador de textura TA-XT2), fração de derretimento, overrun (durante a elaboração do produto) e perfil lipídico. Para o teste de aceitabilidade do produto, utilizou-se uma escala hedônica estruturada de 9 pontos. Elevada viabilidade probiótica foi observada para todas as formulações, com médias de populações acima de 8,0 log UFC/g, não diferindo significativamente durante o armazenamento de 112 dias (p>0,05). B. animalis subsp. lactis HN019 apresentou uma maior resistência em relação a L. acidophilus NCFM quando submetido aos sucos gastrointestinais artificiais, uma vez que a população de NCFM e de HN019 diminuíram, respectivamente, cerca de 5,2 log UFC/g e de 1,2 log UFC/g, durante o armazenamento. O efeito protetor do WPC e/ou I sobre a resistência de L. acidophilus aos sucos gastrointestinais artificiais foi observada no 56º dia e, para B. animalis subsp. lactis no 2º dia de armazenamento (p<0,05). Os sorvetes com WPC apresentaram menores valores de dureza, aos 7º e 112º dias de estocagem (p<0,05). A adição de inulina influenciou no aumento da dureza para F2 após 56 dias e para F4 durante todo período de armazenamento (p<0,05). Os resultados mostraram, também, que a presença do WPC e/ou inulina reduziu a velocidade de derretimento dos sorvetes durante todo o armazenamento (p<0,05). Elevados escores médios (entre 6,8 e 8,0) foram obtidos no teste de aceitabilidade sensorial dos sorvetes probióticos, indicando excelente aceitação pelos consumidores e não diferiram significativamente durante o armazenamento de até 84 dias. Já para F4, a adição do WPC + I aumentou a aceitação do produto após 56 dias (p<0,05). Os resultados obtidos sugerem que a utilização do WPC como substituto parcial da gordura láctea separadamente ou combinada com a inulina pode ser vantajosa no desenvolvimento de sorvete probiótico com baixo teor de gordura, uma vez que a presença desses ingredientes desempenhou um papel importante na proteção dos probióticos contra o efeito dos fluidos gastrointestinais nos testes in vitro. Além deste efeito protetor, a utilização da inulina e WPC também melhorou as características tecnológicas e sensoriais do sorvete funcional reduzido de gordura


This study aimed to assess the effect of the addition of inulin (I) and the partial substitution of the milk fat (MF) by whey protein concentrate (WPC) on Lactobacillus acidophilus NCFM and Bifidobacterium animalis subsp. lactis HN019 viability incorporated in low fat graviola (Annona muricata L.) ice-cream and on probiotic survival under in vitro simulated gastrointestinal conditions throughout 112 days of storage. Moreover, the influence of these ingredients (6% I; 1,5% WPC; 3% and 1,5% MF) on the functional ice-cream technological and sensorial features was also evaluated. Employing a 22 factorial design, four formulations were produced, in triplicates: F1- control (- I, - WPC); F2 (+ I, - WPC); F3 (- I, + WPC) and F4 (+ I, + WPC). The product were stored at -18±3ºC and analyzed after 2, 28, 56, 84, and 112 days of storage. Ice-creams from each trial were used for determination of L. acidophilus and B. animalis subsp. lactis viability in the products and survival in ice-creams submitted to gastrointestinal simulated conditions during storage at -18±3ºC for up to 112 days. For the determination of technological features, instrumental hardness (in TA-XT2 Texture Analyser), melting rate, overrun (during production), and lipid profile were determined. For sensory acceptability evaluation, a 9 point hedonic scale was used. High probiotic viability was observed for all formulations, with mean populations above 8.0 cfu/g and which did not differ significantly throughout 112 days of storage (p>0.05). B. animalis subsp. lactis HN019 resistance to the artificial gastrointestinal juices was higher than for L. acidophilus NCFM, since the NCFM and the HN019 populations decreased approximately 5.2 log cfu/g and 1.2 log cfu/g, respectively, throughout storage. The protective effect of WPC and/or WPC + I on the L. acidophilus resistance to artificial gastrointestinal juices was observed on the 56th day and for B. animalis subsp. lactis on the 2nd day of storage (p<0.05). The ice-creams with WPC presented lower hardness in the 7th and 112nd days of frozen storage (p<0.05). The addition of inulin led to an incresed hardnes for F2 after 56 days and for F4 during the whole storage (p<0.05). The results also showed that the presence of the WPC and/or inulin reduced the ice-creams melting rates during the whole storage (p<0.05). The high mean scores obtained (between 6.8 and 8.0) in the acceptability test indicated that the functional ice-creams evaluated were very well accepted, and did not differ significantly throughout storage of up to 84 days. Except for F4, the addition of the WPC + I improved the acceptability after 56th days of frozen storage (p<0.05). The results suggest that the use of WPC for the partial substitution of the milk fat separately or combined with inulin may be advantageous in the development of low-fat synbiotic ice-cream, since the presence of these ingredients played an important role in the probiotic protection against gastrointestinal juices in the in vitro simulated assays. Besides these protective effects, inulin and WPC also improved the technological and sensory features of the low-fat functional ice-cream

ABSTRACT

O uso do soro lácteo como complemento na formulação de alimentos visando ao seu aproveitamento é uma alternativa para a obtenção de novos produtos. Neste trabalho, desenvolveram-se bebidas à base de soro oriundo de queijo coalho tipo A com frutas e hortaliças. Na formulação das bebidas, utilizaram-se diferentes proporções entre soro lácteo, frutas e hortaliças, com adição de 10% de açúcar. Seis bebidas foram formuladas e submetidas a um teste de preferência, e as três primeiras colocadas foram submetidas a um teste de aceitação sensorial. As três preferidas foram as bebidas sabor graviola, morango e goiaba. A bebida sabor graviola obteve aceitação com as melhores notas nos atributos sensoriais cor, sabor, aparência e qualidade global, seguida pela formulação de sabor morango, mas sem diferença significativa (P>0,05). A bebida sabor goiaba teve o diferencial no atributo odor e superou as bebidas sabor graviola e morango (P<0,05). Conclui-se que é viável a elaboração de bebidas à base de soro de queijo coalho com frutas.


The use of whey as complement in food formulation aiming it's an alternative for the attainment of new products. The aim of this work was developed a type A curd cheese whey derived drink with fruits and vegetables. On drink's formulation was used different ratios of whey, fruits and vegetables, with 10% of sugar. Six drinks were formulated and submited to a preference test. The three best drinks were submitted to a acceptance test. The three best drinks were graviola, strawberry and guava flavours. The graviola one had the greater acceptability with best grades in color's sensory attributes, flavor, appearance and overall quality, following the strawberry flavor, but without significant difference (P<0,05). The guava dink had it's differential on scent character, overcoming the graviola and strawberry drinks (P<0,05). In conclusion we can say that whey-based drink with fruits is viable.

ABSTRACT

Insects collected on soursop (Annonaceae: Annona muricata L.) nearby Manaus, Amazonas, Brazil, are reported. Thirty-seven species were found, but only six were considered injurious to the plant: Bephratelloides pomorum V. (Eurytomidae), Cerconota anonella Sepp (Stenomatidae), Membracis suctifructus Boulard & Couturier (Membracidade) and Pinnaspis aspidistrae Signoret (Diaspididae) attacking fruits; Aphis spiraecola Patch and Aphis gossypii Glover (Aphididae) feeding on leaves, and Cratosomus bombina F. (Curculionidae) boring trunks and branches. Twenty four species are recorded for the first time.


Os insetos da graviola (Annonaceae: Annona muricata L.) foram estudados na região de Manaus, Amazonas, Brasil. Encontram-se 37 espécies, entre os quais, seis foram consideradas prejudiciais, quatro atacando o fruto (Bephratelloides pomorum F. (Eurytomidae), Cerconota anonella Sepp (Stenomatidae), Membracis suctifructus Boulard & Couturier (Membracidae) e Pinnaspis aspidistrae Signoret (Diaspididae); duas nas folhas jovens; (Aphis spiraecola Patch e A. gossypii Glover (Aphididae); e uma perfurando tronco e ramos, Cratosomus bombina (Curculionidae). Trinta e quatro espécies são registradas pela primeira vez.
